Calprotectin is a small protein, normally comprised of two subunits.  The protein is produced in abundant amounts in neutrophil cells, normally about 45% of the total cytosolic protein in the cells. The protein is also produced in many other types of cells. Calprotectin plays several roles in the human immune system. Perhaps the most important role is as an early signal when a pathogen is being detected and targeted, activating innate immune cells and allowing the orchestration of an inflammatory response. 

In relation to Lyfstone® ’s use of Calprotectin as a biomarker, this happens when neutrophil cells swarm into an infected joint. The cells activate and burst, releasing large amounts of pre-produced Calprotectin into the synovial fluid. This means that the signal the Lyfstone® test detects is clear and strong, and well-connected to the clinical issue at hand.

The letters stand for Periprosthetic Joint Infection. The word Periprosthetic in medicine refers to a structure in close relation to an implant, in this case, an artificial joint. There have been many attempts to define the criteria by which PJI is diagnosed. However, no single definition has gained acceptance as the reference standard for clinical practice.

Any diagnostic definition set has two distinct uses: firstly, and most importantly, it must give a clinician, who may or may not be an expert in the field, a clear set of investigations and outcomes which allow them to decide if an infection is present or not. Secondly, the definition must facilitate researchers to perform studies in patients with or without an infection.

To get started you need:

– A test kit, from one of our national distributors.
– A mobile phone with the Lyfstone® Reader app installed. The app is available for Android and iOS systems.
– A clinic account for using the app/portal system. Please register by opening the Lyfstone® Reader app or at Lyfstone® Portal. 
– Access to laboratory pipettes for 20 µl and 80 µl and aerosol-filter pipette tips.

The Lyfstone® Reader app to scan the Lyfstone® Calprotectin Test can be downloaded from the App Store for iOS or Play store for Android devices. Please make sure to install the ‘Lyfstone® Reader’ app.

No. Download the app on all devices to be used, and log in with the same username and password.

The app takes a normal picture of the test in the frame when the lines on the screen align with the test and frame below. The app then uses the pre-programmed set-up of the test in the frame to select two regions of interest. One covers the control line, the other the test line. These lines are analysed and quantified using the algorithm programmed in the app. The finished result is given based on the ratio of test line reading over control line reading. The ratio is a more robust result than just the reading of one or the other lines, given that most factors that influence the expressions and reading of the lines influence both simultaneously.

The test is NOT developed to be read without the reader and is not legally/regulatory approved for such use.

The control and test lines should be clear and well-defined. If the lines are missing or the quality of the lines is not acceptable, the software will be unable to detect the test line, and/or an error message will appear. A new test should be performed if this occurs. If the QR code is damaged, the cassette will not be detected by the software and an error message will appear. Please also double-check on the box if the test is not expired.
